Splice opens with a moment of ominous creation, The viewer knows this based on the background music playing over the opening credits (a series of veins bulging across pale skin to spell out cast and crew). In a Toronto lab, genetic engineers Clive Nicoli (Adrien Brody, who looks physically wounded at having been cast) and Elsa Kast (Sarah Polley, seemingly wondering if this role will kill her career) act as midwives to a hybrid lifeform (dubbed "Ginger") looking like a cross between the monstrous puddlinglike offspring in David Lynch's Eraserhead and a phallic tumor from David Cronenberg's The Brood.
Synopsis
Superstar genetic engineers Clive Nicoli (Adrien Brody) and Elsa Kast (Sarah Polley) have achieved fame by successfully splicing together the DNA of different animals to create incredible new hybrid animals, including two dog sized worms. Now they want to use human DNA in a hybrid that could revolutionize science and medicine. But when the pharmaceutical company that funds their research, N.E.R.D., forbids it, Clive and Elsa decide to secretly conduct their own experiments in the pursuit of even greater scientific discovery. They secretly blend human DNA with that of their creations, ignoring their society's ethical and legal boundaries.
The scientists name their creature Dren (nerd spelled backward) and at first she exceeds their wildest dreams; growing and learning at an accelerated rate. As their lab becomes exceedingly crowded, and the risk of Dren being discovered increases, they move her to Elsa's dead mother's farm. Their work suffers due to the amount of time spent taking care of Dren, and their colleagues become hostile; Clive's brother even discovers Dren, and is barely made to keep silent. Clive's conscience constantly drives him to fight the experiment, even to the point where he tries to drown Dren (though her amphibious lungs render the attempt unsuccessful).
Meanwhile, the dog like worms that they had created (which were originally mates) kill each other after the female became a male and extremely hostile. Later on, Clive discovers that Elsa used her own DNA to create Dren, which by now has become amphibious, winged and omnivorous with a neurotoxic sting on her tail. Elsa dotes on her, considering her a child of sorts, and tries to humanize her by clothing her in a dress, putting makeup on her, and sharing her secret childhood toys with the creature. Dren ages very quickly, progressing from an innocent childlike stage to an intelligent teenager who can communicate by arranging Scrabble tiles into words (she has no ability to speak). After a time, Dren quickly grows bored with hiding inside Elsa's barn and becomes frustrated that Elsa won't let her have what she wants, including a stray cat she discovered inside the barn. Tensions between the two women flare until Elsa realizes that she is turning into her own distant and oppressive mother, a harpy who never let her have any toys and forced her to live in a barren room. Elsa tries to make amends by bringing the cat back, but Dren stings and kills the cat, then attacks Elsa. Elsa manages to break free and knock her out with a shovel. She then decides that she has gotten too close to Dren and now treats her simply like an experiment, cutting off the creature's stinger and stripping her of her clothes and makeup. From the stinger, she isolates proteins that are far beyond the worms' results, tipping off the company that human DNA is present in the sample she used.
When Dren is freed, she seduces and mates with Clive, much to Elsa's disgust and then it is shown that her stinger has regenerated. The two "parents" decide that they have been creating a parody of a child, and now is the time to end their out of control experiment by killing Dren. However, when they arrive at the farm, they discover that she is already dying in her water tank inside the barn. Elsa and Clive watch her die, then bury her next to the barn. They are just about to leave when the pharmaceutical company head scientist and Clive's brother arrive, demanding to see Dren. When they go to see her body, Dren attacks them, revealing that she has become a territorial male just like the original hybrid animals. Dren overpowers and rapes Elsa, then kills Clive with a toxic sting. Dren finally dies when Elsa smashes his head with a rock. Later on, Elsa is seen in the office of the pharmaceutical company's head and is given a large amount of money in exchange for her silence and taking the experiment to the next stage. The head of the company then thanks Elsa for the personal risk that she is undergoing. Elsa stands up and is revealed to be pregnant, presumably with Dren's child.
